چکیده

Unilever initiated a long-term research and capability development program in 2004 to evaluate and implement a new conceptual approach for assuring consumer safety without animal testing (Fentem et al., 2004, Westmoreland et al., 2010). Unilever’s approach for safety assessment is risk-based, meaning that all available data on a new ingredient (including predicted levels of consumer exposure during product use) are used to assess the level of risk posed by its proposed use. The scientific challenge we continue to investigate is how novel non-animal datasets (i.e., data generated using in chemico and in vitro test methods) can be applied to risk assessment decision making to replace the need for new animal test data. Within our Skin Sensitization program we have made a long-term investment to define and explore the key toxicity pathways that drive the disease process (i.e., Allergic Contact Dermatitis) in humans, as we believe this mechanistic understanding should form a sound basis for our non-animal risk assessment approach (Maxwell et al., 2008). Here we share our progress in developing a mechanisticallybased mathematical model of the induction of skin sensitization in humans.

From pathways to people: applying the adverse outcome pathway (AOP) for skin sensitization to risk assessment.

Consumer safety risk assessment of skin sensitization requires information on both consumer exposure to the ingredient through product use and the hazardous properties of the ingredient. Significant progress has been made in determining the hazard potential of ingredients without animal testing. However, hazard identification is insufficient for risk assessment, and an understanding of the dose...

Predicting chemically-induced skin reactions. Part II: QSAR models of skin permeability and the relationships between skin permeability and skin sensitization

Skin permeability is widely considered to be mechanistically implicated in chemically-induced skin sensitization. Although many chemicals have been identified as skin sensitizers, there have been very few reports analyzing the relationships between molecular structure and skin permeability of sensitizers and non-sensitizers. The goals of this study were to: (i) compile, curate, and integrate th...

Safety Evaluation of Cosmetic Ingredients Regarding Their Skin Sensitization Potential

Up to today, product safety evaluation in the EU is predominantly based on data/information on their individual ingredients. Consequently, the quality and reliability of individual ingredient data is of vital interest. In this context, the knowledge about skin sensitization potential is an explicit need for both hazard and risk assessment. Proper skin sensitization data of the individual chemic...

OECD GUIDELINE FOR THE TESTING OF CHEMICALS DRAFT PROPOSAL FOR A NEW TEST GUIDELINE In Vitro Skin Sensitization: Keratinocyte-based ARE-Nrf2 Luciferase Reporter Gene Test Method

1. A skin sensitizer refers to a substance that will lead to an allergic response following skin contact (1). There is general agreement regarding the key biological events underlying skin sensitization summarized as an Adverse Outcome Pathway (AOP) which includes four events recognized as key ones, going from the molecular initiating event to the adverse health effect, i.e., allergic contact d...

Predictive identification of human skin sensitization thresholds.

For years, methods have been available for the predictive identification of chemicals that possess the intrinsic potential to cause skin sensitization. However, many have proven less suitable for the determination of relative sensitizing potency. In this respect, the local lymph node assay (LLNA) has been shown to have a number of important advantages. Through interpolation of LLNA dose-respons...
